Reclassification of Streptomyces caeruleus as a synonym of Actinoalloteichus cyanogriseus and reclassification of Streptomyces spheroides and Streptomyces laceyi as later synonyms of Streptomyces niveus.

Previous studies have proposed that Streptomyces caeruleus is an earlier heterotypic synonym for Streptomyces niveus and Streptomyces spheroides. In this study, phylogenetic analysis of the almost complete 16S rRNA gene sequences of the Streptomyces caeruleus type strains NBRC 13344(T), JCM 4014(T) and NRRL B-2194(T) revealed that S. caeruleus was closely related to Actinoalloteichus cyanogriseus and not to S. niveus, S. spheroides or any other species of the genus Streptomyces. Moreover, the diagnostic cell-wall diamino acid was found to be meso-diaminopimelic acid in S. caeruleus and DNA-DNA hybridization studies revealed that S. caeruleus NBRC 13344(T) was a member of the same species as A. cyanogriseus NBRC 14455(T). Based on these chemotaxonomic and phylogenetic data, it is proposed that Streptomyces caeruleus (Baldacci 1944) Pridham et al. 1958 be reclassified as a heterotypic synonym of Actinalloteichus cyanogriseus Tamura et al. 2000. Furthermore, based on phylogenetic, morphological and MALDI-TOF MS analyses, it is proposed that the species Streptomyces laceyi Manfio et al. 2004 and Streptomyces spheroides Wallick et al. 1956 are reclassified as later heterotypic synonyms of Streptomyces niveus Smith et al. 1956.

Phylogenetic analysis of 'Actinoalloteichus cyanogriseus' based on its 16S rDNA sequence confirmed that the organism belongs to the family Pseudonocardiaceae. It contains glutamic acid, alanine and meso-diaminopimelic acid as cell wall amino acids, and menaquinone 9 (H4). Its taxonomic characteristics differ from those of the previously described organisms of the family Pseudonocardiaceae. On the basis of these morphological, physiological, chemotaxonomic and phylogenetic analyses, a new genus, Actinoalloteichus, is proposed, along with a new species, namely Actinoalloteichus cyanogriseus sp. nov. The type strain is A. cyanogriseus IFO 14455T (= AS 4.1159T = JCM 6095T).

A collection of 93 Streptomyces reference strains were investigated using SDS-PAGE of whole-cell proteins. Computer-assisted numerical analysis revealed 24 clusters encompassing strains with very similar protein profiles. Five of them grouped several type strains with visually identical patterns. DNA-DNA hybridizations revealed homology values higher than 70% among these type strains. According to the current species concept, it is proposed that Streptomyces albosporeus subsp. albosporeus LMG 19403T is considered as a subjective synonym of Streptomyces aurantiacus LMG 19358T, that Streptomyces aminophilus LMG 19319T is considered as a subjective synonym of Streptomyces cacaoi subsp. cacaoi LMG 19320T, that Streptomyces niveus LMG 19395T and Streptomyces spheroides LMG 19392T are considered as subjective synonyms of Streptomyces caeruleus LMG 19399T, and that Streptomyces violatus LMG 19397T is considered as a subjective synonym of Streptomyces violaceus LMG 19360T.
